Synergistic induction of HLA class I expression by RelA and CIITA.

چکیده

The major histocompatibility complex (MHC) class I genes are induced synergistically by interferons (IFN) and tumor necrosis factor (TNF), a response thought to involve the cooperative action of Rel/NF-kB and interferon regulatory factor (IRF) transcription factors. The IFN-gamma-inducible class II transcriptional activator (CIITA) has recently been shown to transactivate MHC class I as well as class II genes, and this investigation shows that CIITA synergizes strongly with RelA to stimulate HLA class I expression. The functional interaction of CIITA and RelA requires both promoter elements and the upstream Rel binding site and is not seen with a class II reporter. The promoter elements necessary for CIITA action are also required for induction by IFN-alpha. HLA-A and HLA-B loci respond differentially to IFNs, and we identify locus-specific differences in critical promoter elements in addition to known polymorphisms in the Rel and IRF binding sites. The HLA-A promoter is transactivated relatively poorly by CIITA and does not interact detectably with CREB proteins implicated in CIITA recruitment, but the synergism with RelA can compensate for this weakness. The present findings illustrate that multiple transcription factors cooperate to regulate class I expression and that their relative importance differs according to the locus and cell type examined. CIITA enhances HIV-1 attachment to CD4+ T cells leading to enhanced infection and cell depletion.

Activated CD4(+) T cells are more susceptible to HIV infection than resting T cells; the reason for this remains unresolved. Induction of CIITA and subsequent expression of the MHC class II isotype HLA-DR are hallmarks of CD4(+) T cell activation; therefore, we investigated the role of CIITA expression in T cells during HIV infection. CIITA-expressing SupT1 cells display enhanced virion attachm...

Reports from the 15th European Histocompatibility Conference

and Poster Viewing Sessions Thorsten Zacher, Erlangen, Germany The first set of reported abstracts gave an overview of the current progress in understanding the regulation of the transcription factor CIITA, focusing on DNA methylation as a way of silencing HLA class II but also class I gene expression. In the second set of abstracts I concentrate on all presented abstracts dealing with HLA anti...

Class II transactivator is required for maximal expression of HLA-DOB in B cells.

HLA-DO, encoded by the HLA-DOA and HLA-DOB genes, has been shown to function as a modulator of Ag presentation. DNA microarray comparisons between B cells wild-type and mutant for the master regulator of MHC class II transcription, class II transactivator (CIITA), identified HLA-DOA and HLA-DOB as being up-regulated by CIITA. Although HLA-DOA had been shown previously to be regulated by CIITA, ...

بررسی رابطه بین ظاهر شدن مولکولهای کلاس HLA-I بر سطح یاخته‌های رده سرطانی و حساسیت به سلولهای کشنده طبیعی

ABSTRACT Natural Killer cells (NKs), are considered an important first line of defence and were originally defined by their ability to kill certain tumor cells and virally infected cells. Our aim is to study expression of HLA class-Ia on tumor cell lines and their susceptibility of NK cells. In this study MDA-MB-468 tumor cell line (NCB1, C208) That derived from human breast adenocarcino...

Can the Decreased Expression of Human Leukocyte Antigen Class Ⅰ and Ⅱ by Spermatozoa Lead to Recurrent Spontaneous Abortion?

Background & Objective: Unexplained recurrent spontaneous abortion (URSA) is defined as an unknown cause of occurrence of three or more clinically detectable pregnancy losses before 20 weeks of gestation, but it occurs presumably as a result of the immune system dysfunctions. We supposed that the disruption of semen or spermatozoa might be responsible for the dysfunction of the...
